Possibilities:

• Defeating 4 opponents gets you across the floor, but does not gaurantee it immediately (draws count). Test #7 weakens this theory considerably.
• A random # for kills needed is chosen each time you try to get through, in the range of 4-5. Odd choice, but I suppose possible.
• You need a kill counter of X to get across the floor; each opponent adds a certain # to your kill counter. Running away, draws, and being defeated may all affect it somehow (or may not). Reaching the required kill # does not gaurantee an immediate stairway. Note that test 7 makes it seem very unlikely that running away makes you lose progress. (Funny thought: what if there are two, identical or near-identical 'etheric' and 'non-etheric' writhers. I don't know why this would be the case, unless they behaved differently for the kill counter. Um, yeah, weird thought, forget it).
• There is no kill counter, getting to the stairs is just random chance (unlikely considering test #7 and other unrecordered attempts)
• Defeating opponents increases the chances of the stairs appearing, by a certain % for each opponent.

Testing strongly implies that there's not just a 3-kill counter here. I killed 2 enemies, lost to another, then killed a 3rd, and got enemies (running away) the next several times. Either scientists don't count as much, or being defeated requires you to fight some more, or there's a random kill counter chosen. Hm… defeating another scientist and running away didn't help matters. After killing a hound, I immediately got the stairs. Wish I had recorded this…

Okay, test #9 suggests that 4 scientists are required to get across the floor if that's all you kill (and if you don't lose any fights). There's also the possibility that there's a random # chosen (sometimes 3, sometimes 4) and enemy type doesn't matter. I find that unlikely, but it's worth trying the 3-scientist thing again sometime.

Completely off topic, I wasn't expecting this to be a 2 day run. Shows how much I know. :P

(previously ignored and now beloved energy sources: protein bars and Nova, the latter right before you enter Biotech - running away only uses 1 energy, as usual)

Okay, seems pretty obvious that you need to get through the roads encounter, then the stairs are free, though not an immediate guarantee (but with high probability, perhaps due to their being only one other encounter).

(#5: high road, stairs) (finished 2-dayer with best margin yet: 16 energy and 8 body, with 4 blitz poppers ready if need be. Stuffing my face with protein bars helped)
(#6: eye, eye, captain, high road, stairs… minus the captain) (Autodoc + reinforced surgical mask healed me for 21 in those 3 combats alone, heh heh) (another 2-dayer, with 9 energy and 7 body left. Protein bars to the rescue once again)
(#7: eye, eye, high road, bonny lakes, eye, stairs… minus the bonny lakes)(I passed on a possible 2-dayer yesterday due to real life, so this is a 3-dayer. Which gave me a lot of time to do interesting spading here. :) )

Note that these numbers don't correspond cross-floors. Test #7 for the fifth floor may have been #6, #8, or an unrecorded fourth floor, for instance.

I passed the third floor in 3 turns:

Got to the third floor
Got the acid rain adventure (in containment suit)
Got the acid rain adventure (in containment suit)
Found the stairs to the fourth floor

If there's any requirement for this floor, this might narrow it down a bit.

I got the same thing. Two turns of acid rain, with only a containment helmet and then stairs.
